Description

Principle of the Assay: Mouse R-Spondin-3 ELISA Kit was based on standard sandwich enzyme-linked immunesorbent assay technology. A monoclonal antibody from mouse specific for R-Spondin-3 has been precoated onto 96-well plates. Standards(Expression system for standard: NSO; Immunogen sequence: Q22-H277) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for R-Spondin-3 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Mouse R-Spondin-3 amount of sample captured in plate.

Background/Introduction: R-spondin-3 is a protein that in humans is encoded by the RSPO3 gene. This gene belongs to the Rspondin family. The encoded protein plays a role in the regulation of Wnt (wingless-type MMTV integration site family)/beta-catenin and Wnt/planar cell polarity (PCP) signaling pathways, which are involved in development, cell growth and disease pathogenesis. Genome-wide association studies suggest a correlation of this gene with bone mineral density and risk of fracture. This gene may be involved in tumor development
